Abstract

Background: Pediatric rare diseases (PRDs) affect millions of children worldwide and are often associated with severe genetic abnormalities and progressive disability with few treatment options. The usual treatments are directed toward controlling symptoms rather than toward removing the basic causes of disease. “New possibilities have been opened up to explore curative and personalized treatment approaches through recent advances in gene editing and regenerative medicine technologies.  Objective: To assess the efficacy of new gene editing and regenerative medicine technologies for treating rare pediatric diseases and improving clinical outcomes. Methodology: An in-depth study was performed through clinical studies, genomic data sets, and regenerative medicine reports. Therapeutic approaches studied were CRISPR-Cas9 gene editing, base editing, stem cell therapy, induced pluripotent stem cells (iPSCs), tissue engineering and regenerative medicine-based interventions. Treatment efficacy, rates of genetic correction, tissue regeneration outcomes, and patient recovery indicators were assessed.  Findings: Genetic correction accuracies of up to 95.1% were achieved with gene editing technologies, and tissue repair and functional recovery were improved by approximately 88.6% with regenerative medicine approaches. The combination of therapies improved clinical outcomes by 41% and decreased the rate of disease progression by almost 36% compared to traditional therapies.  Conclusion: Gene editing and regenerative medicine approaches have promise for therapeutic approaches for rare diseases in childhood. The ability to target genetic defects and to promote tissue regeneration offers a significant potential for personalized treatment, improved quality of life and long-term disease management.
